Milbemycin Oxime: Veterinary Macrocyclic Lactone Antiparasitic
Milbemycin oxime is a macrocyclic lactone derived from the fermentation products of Streptomyces hygroscopicus subsp. aureolacrimosus. It acts by opening glutamate-gated chloride channels in invertebrate nerve and muscle cells, causing hyperpolarization, paralysis, and death of susceptible parasites. It does not affect mammalian chloride channels at therapeutic doses.

Applications
Milbemycin oxime is widely used in dogs and cats for the prevention of heartworm disease (Dirofilaria immitis) and the treatment of intestinal nematode infections including roundworms and hookworms. In dogs, it is also effective against whipworm (Trichuris vulpis). It is commonly formulated in combination with praziquantel for broad-spectrum coverage against both nematodes and cestodes. Milbemycin oxime is available as oral tablets and chewable formulations. Unlike ivermectin, milbemycin oxime has a wider safety margin in collie breeds with the MDR1 gene mutation, though caution is still advised at higher doses.
